We conclude this Review with a few suggestions as guides for future work.It is widely accepted that the pathogen Clostridioides difficile exploits an intestinal environment with an altered microbiota, but the details of these microbe-microbe interactions are unclear. Adherence and colonization of mucus has been demonstrated for several enteric pathogens and it is possible that mucin-associated microbes may be working in concert with C. difficile. We showed that C. difficile ribotype-027 adheres to MUC2 glycans and using fecal bioreactors, we identified that C. difficile associates with several mucin-degrading microbes. C. difficile was found to chemotax toward intestinal mucus and its glycan components, demonstrating that C. difficile senses the mucus layer. Although C. difficile lacks the glycosyl hydrolases required to degrade mucin glycans, coculturing C. difficile with the mucin-degrading Akkermansia muciniphila, Bacteroides thetaiotaomicron, and Ruminococcus torques allowed C. difficile to grow in media that lacked glucose but contained purified MUC2. Collectively, these studies expand our knowledge on how intestinal microbes support C. difficile.Nanospray desorption electrospray ionization (nano-DESI) has been utilized in direct sampling mass spectrometry (MS) that requires highly spatially resolved sampling with minimal sample destruction. In this study, we explored the applicability of nano-DESI MS for the forensic chemical analysis of ink directly from handwriting on paper. Nano-DESI readily ionizes dyes, including the polyanionic ones, with minimal fragmentation and produces chemical fingerprints of ballpoint pens directly from a paper surface. Further, we specifically focused on how the potential of nano-DESI that changes the mass spectral profiles over time could reflect the differential distribution of analytes in a vertical direction because mildly extracted analytes are immediately transferred and analyzed in real time. To test this, we wrote the character "X" with various combinations of two different pens and analyzed the crosspoints by nano-DESI MS. As a result, the time-course changes in the chemical fingerprints of the ink, which were consistent with the order of the pen strokes, were successfully obtained by nano-DESI MS in most cases. Both equilibrium (EQ) and nonequilibrium (NE) molecular dynamics (MD) simulations are employed with chemically specific coarse-grained models. The fluctuation equation seems to be a simple route to calculate the B, but the result can significantly depend upon the barostat mass. The thermodynamics relation is valuable to disclosing the entropy contribution, but the result can suffer from great errors in nonlinear fitting. In particular, the instantaneous pressures and volumes during the EQ MD simulations in the "true" NPT ensemble are thus proposed to calculate the B via the definition formula. It is demonstrated in both case-by-case and averaged ways that the effects of tacticity on the B of the polymer are very slight. Recently, microRNAs (miRNAs) have been reported to participate in PE progression, whereas the precise functions and potential mechanisms of miR-20b in placental trophoblast cells as well as in PE progression remain poorly understood. In the present study, real-time quantitative polymerase chain reaction (RT-qPCR) analysis was used to detect expressions of miR-20b and myeloid cell leukemin- 1(MCL-1) mRNA. Cell viability was investigated by cell counting kit-8(CCK-8) assays. Cell invasion and migration abilities were determined by Transwell assays. Western blot was performed to detect MCL-1 protein expressions. The interaction between miR-20b and MCL-1 was investigated by bioinformatics analysis and luciferase activity assay. The results of the study demonstrated that miR-20b was highly expressed in placental tissues of patients with PE. Moreover, miR-20b overexpression inhibited HTR8/ SVneo cell proliferation, invasion and migration. Furthermore, MCL-1 was targeted by miR-20b, and MCL-1 restoration could partially attenuate the effect of miR-20b on HTR8/SVneo cells. In conclusion, the results indicate that miR-20b may contribute to PE through inhibiting proliferation, invasion and migration of placental trophoblast cells by targeting MCL-1. Therefore, miR-20b may be used as a notable biomarker for the diagnosis, prevention, and treatment of PE.
